T-cell immunoreceptor with Ig and ITIM domains is an immunoglobulin superfamily receptor encoded by the TIGIT gene. It is a type I single-pass transmembrane glycoprotein with an extracellular immunoglobulin variable-like domain and a cytoplasmic tail containing immunoreceptor tyrosine-based inhibitory motifs. TIGIT is expressed mainly on activated T cells, regulatory T cells, natural killer cells, and subsets of memory lymphocytes, where binding to ligands such as PVR (CD155) and NECTIN2 (CD112) modulates immune cell activation and effector responses. Its intracellular signaling is associated with inhibitory regulation of cytokine production and cytotoxic activity.
