Glucagon-like peptide-1 receptor is a member of the class B secretin family of G protein-coupled receptors encoded by the GLP1R gene. It is a seven-transmembrane cell surface receptor with a large extracellular N-terminal ligand-binding domain that recognizes glucagon-like peptide-1 and related agonists. GLP1R primarily couples to Gs proteins to stimulate adenylyl cyclase and increase cyclic AMP, leading to enhanced glucose-dependent insulin secretion and modulation of cellular excitability. It is expressed prominently in pancreatic islet beta cells and in selected tissues including gastrointestinal tract, kidney, heart, and regions of the nervous system.
