Claudin-4 is a member of the claudin family of tight junction proteins encoded by the CLDN4 gene. It is an integral membrane protein with four transmembrane helices, two extracellular loops, and cytoplasmic N- and C-terminal tails that participate in tight junction strand formation and paracellular barrier regulation. Claudin-4 is expressed in multiple epithelial tissues, including kidney, lung, and gastrointestinal tract, where it contributes to maintenance of cell polarity and selective permeability between adjacent cells. Its extracellular domains mediate homotypic and heterotypic interactions with other claudins within tight junction complexes.
