Claudin-2 is a member of the claudin family of tight junction proteins encoded by the CLDN2 gene. It is an integral membrane protein with four transmembrane helices, two extracellular loops, and cytoplasmic N- and C-terminal tails that participate in tight junction strand assembly. Claudin-2 is expressed in epithelial tissues including kidney and intestine, where it forms cation-selective paracellular channels that influence permeability to small ions and water. Its distribution contributes to regulation of epithelial barrier properties and transepithelial transport.
