Claudin-1 is a member of the claudin family of tight junction proteins encoded by the CLDN1 gene. It is an integral membrane protein with four transmembrane helices, two extracellular loops, and cytoplasmic N- and C-terminal tails that contribute to tight junction strand formation. Claudin-1 is expressed in epithelial and endothelial tissues where it regulates paracellular barrier properties, cell polarity, and intercellular adhesion. It is particularly abundant in skin and other barrier-forming epithelia and is required for normal epidermal barrier function.
